NMN and NR
What is nmn and nr?
NMN (nicotinamide mononucleotide) and NR (nicotinamide riboside) are forms of vitamin B3 that the body turns into NAD+, a molecule cells need to make energy and repair damage. NAD+ falls with age in animals, so some people take these supplements to try to slow aging. They raise NAD+ in human blood, but no trial has shown they slow aging.
Listed here because well-known people have tried it on themselves. Trying something is not recommending it; each entry says whether the person tells others to do it.

Kept
David SinclairGeneticist at Harvard Medical School, author of Lifespan
Dates not stated · Does not recommend it to others
Takes 1 g of NMN every morning and has for more than a decade, as part of a protocol built on his lab's research on NAD+ and aging.

Andrew HubermanNeuroscientist at Stanford, host of Huberman Lab
Dates not stated · Does not recommend it to others
Takes 1.5 g of NMN under the tongue each morning and has also taken NR; he likes the energy he feels but does not see NAD+ boosters as a way to live longer.
“I observe a very clear positive effect, but I don't think it has anything to do with extending lifespan.”

Dates not stated · Does not recommend it to others
Lists NMN, an NAD+ precursor, among the supplements he takes every day.
“Here is a list of the meds and supplements I take each day to help extend my longevity and healthspan”

Charles BrennerBiochemist at the University of Helsinki, chief scientific advisor of Niagen Bioscience
Dates not stated · Recommends it to others
Takes NR as Niagen each morning; he discovered NR as an NAD precursor, advises Niagen Bioscience, which sells it, and calls 500 to 1,000 mg a substantive daily amount.
“I'm on team breakfast I'm not a time restricted guy I take Niagen in the morning I have coffee and I work” (47:41)

What the evidence shows
In people, both raise NAD+. In a six-week crossover trial in healthy middle-aged and older adults, NR was well tolerated and raised NAD+ metabolism in blood [1]. In a 10-week trial, NMN improved muscle insulin sensitivity in postmenopausal women with prediabetes and excess weight [2]. A 12-week trial of a high dose of NR in men with obesity found no change in insulin sensitivity or body composition [3].
Pooled results are modest. A 2024 meta-analysis of 19 trials found that NAD+ precursors lowered total cholesterol and triglycerides but had no clear effect on other metabolic measures [4], and a 2025 meta-analysis found no benefit for muscle mass or strength in adults over 60 [5]. A 2026 systematic review of 33 human studies concluded that the supplements reliably raise NAD+ markers, while effects on health are mixed and often absent [6].
The striking results are in mice. A year of NMN in normally aging mice limited weight gain and improved energy use, insulin sensitivity and eye function [7]. When the Interventions Testing Program fed NR to genetically mixed mice, it did not extend lifespan [8]. No study has tested whether NMN or NR extends life in people. Neither is an approved drug for any condition; both are sold as supplements.
Safety
- NMN and NR are sold as dietary supplements, not approved drugs. In September 2025 the FDA said NMN may be sold as a supplement in the US, reversing its 2022 position.
- Trials lasting weeks to a few months found both well tolerated. Safety over years of use is unknown.
- The FDA does not check supplements for purity or dose before they are sold, so products vary.
- Whether raising NAD+ for years changes cancer risk has not been studied in people.
- People who are pregnant, breastfeeding or being treated for cancer should talk with a physician before taking them.
Common questions
Does David Sinclair take NMN?
Yes. As of April 2026 he takes 1 g of NMN every morning and has done so for more than a decade.
Does Andrew Huberman take NMN or NR?
Yes. In July 2024 he said he takes 1.5 g of NMN under the tongue each morning and has also taken NR. He likes how he feels on it but does not think NAD+ boosters will extend his life.
Is NMN or NR better?
Do NMN or NR extend lifespan?
Is NMN legal to sell as a supplement?
In the US, yes. The FDA said in 2022 that NMN could not be sold as a supplement because it was being studied as a drug, then reversed that view in September 2025.
